I am sitting with Life, the Universe and everything in front of me. It suggests what the Question (to life, universe, and everything) is, by letting Arthur Dent pulling out random scrabble letters (paraphrasing with the pure intent to make things shorter. The context is still the same):
"What do you get if you multiply six by nine..." And then he runs out of letters.
Another theory about the question, which was suggested by the intelligent mice in Hitchiker's Guide to the Galaxy itself was "How many roads must a man walk down?". This is commonly known as the cheap way out, for sounding significant without having any meaning. Or as the other mouse said "Frankie baby, we are made!
EDIT! EDIT! EDIT! I did a major mistake. Arthur Dent did of course get close to asking the question in Chapter 33 in The Restaurant At the End of the Universe. To my defence, my copy of the two books is (together with HGttG and So Long, and THanks for All the Fish) simply one big volume.

Rubbish, though. This was the response created by the Earth Mark I whose program had been corrupted by its early colonization by Golgofrinchian exiles.
